New CTMC data on automated, functionally closed formulation and fill-finish with the Xiogenix ARES™ X20+.

As cell therapy manufacturing scales, manual formulation and fill-finish can introduce limitations in throughput, process consistency, and operational efficiency. Addressing these challenges requires downstream solutions that accommodate changing production needs while maintaining critical product quality attributes.

A new white paper from CTMC, developed in collaboration with Xiogenix, evaluates the ARES™ X20+ in tumor-infiltrating lymphocyte (TIL) manufacturing. The functionally closed system automates formulation and filling into cryovials and cryobags, with configurable processing parameters to accommodate different manufacturing workflows.

The study compares automated and manual processing, examining manufacturing efficiency, fill precision, and critical cell quality attributes.

New data on downstream performance

In a CS750 cryobag comparison with CTMC’s standard manual syringe-filling process, automated processing demonstrated:

  • Approximately 40% shorter processing time (58 minutes manual vs. 35 minutes automated)
  • 50% less CS10 exposure time (52 minutes manual vs. 26 minutes automated)
  • Comparable post-formulation cell viability (92.9% manual vs. 93.1% automated), with two operators rather than three

Minimizing DMSO exposure prior to cryopreservation is an important process consideration, particularly given its potential impact on cell viability and post-thaw recovery.

Flexibility as manufacturing needs evolve

The ARES™ X20+ combines configurable recipes, customizable single-use kits, and functionally closed processing to support different formulation volumes and final container formats. CTMC also examined higher-throughput filling, cryovial and cryobag workflows, and post-thaw product quality.

The evaluation also identifies process-optimization considerations that matter when moving from manual operations to a more automated workflow.
